Abstract

Background: Electronic cigarette (vape) use has increased significantly and may cause oxidative stress due to free radical exposure. Bundung leaves (Actinoscirpus grossus) contain secondary metabolites with antioxidant potential. Therefore, they need to be developed into a liquid formulation and evaluated for antioxidant activity. Objective: To determine the antioxidant activity of a liquid formulation containing Actinoscirpus grossus leaf extract using the DPPH method, identify the formulation with the highest antioxidant activity, and evaluate homogeneity and viscosity. Methods: This experimental laboratory study used 250 g of Actinoscirpus grossus leaf powder extracted by maceration with 96% ethanol. The extract was formulated into liquid preparations containing 2.5% (F1), 3% (F2), and 3.5% (F3) extract. Antioxidant activity was determined using the DPPH method with a UV-Visible spectrophotometer, while physical evaluation included homogeneity and viscosity tests. Results: The extraction process produced a yield of 3.86%. Quercetin, the positive control, showed an IC₅₀ value of 2.89 ppm. The IC₅₀ values of F1, F2, and F3 were 380.909, 274.274, and 494.600 ppm, respectively, indicating weak antioxidant activity. F2 demonstrated the highest antioxidant activity. All formulations passed the homogeneity test and exhibited acceptable rheological flow behavior. Statistical analysis showed that variations in extract concentration significantly affected antioxidant activity. Conclusion: The liquid formulation of Actinoscirpus grossus leaf extract exhibited weak antioxidant activity based on IC₅₀ values. F2, containing 1.8 g of extract, was the optimal formulation due to its superior antioxidant activity. All formulations were homogeneous and showed acceptable viscosity characteristics
